1 Design
We performed a large-scale experiment in greenhouse during six months to grow pea plants of two cultivars: Caméor and Kayanne. The Caméor cultivar is represented by 4 genotypes: two mutants obtained by TILLING (affected in the same gene) and their respective wild-type genotypes. So we have 5 different genotypes in this experiment. Just before flowering, we applied 4 different stress conditions for 14 days: no stress, water stress, sulfur deficiency and the two stresses combined. We harvested vegetative leaves at 2 stages: before stress (called E0) and at the end of the stress (named E1) ; and also roots at 3 stages: before stress (E0), at the end of the stress (E1) and during recovery (E2). We aim to : (1) compare responses of these two cultivars to single and double stresses (Kayanne compared to Caméor); (2) study consequences of mutations in the ‘Caméor’ genotype at each stage; (3) compare gene expression across stages for each genotype.
